ABOUT THE CCHMC Genomics Sequencing Facility


The facility is a technical and expertise resource for the genomic research community at Children's Hospital and the University of Cincinnati. We offer a wide variety of services for DNA Sequencing and Genotyping.

We are located in room 1045A of the Research Foundation (R) Building. Our regular hours of operation are 9:00 am to 5:00 pm on weekdays.


  For Sanger Sequencing, we use the ABI PRISM® 3730xl DNA Analyzer, a capillary electrophoresis system developed specifically for production-scale DNA analysis. Sequencing accuracy is > 98.5% to 550 base pairs. Read length can reach over 800 bp with good starting materials. The current capacity is around 1080 sequence reads, or over 10,000 microsatellite-based genotypes per day, 5 days a week.
We have recently purchased the NovaSeq X Plus system from Illumina. This is the current standard bearer in high throughput, short read NGS sequencing. It is capable of running two flow cells concurrently, each containing up to 65 whole-genome libraries at 30x coverage.

 
  The facility is also equipped with two robotic laboratory liquid handlers to automate Sanger sequencing reaction setup, dye terminator removal, Infinium wash and stain procedures for gentoyping assays and other automation.

For long-read sequencing, we utilize the PromethION 2 Solo instrument from Oxford Nanopore Technologies. The PromethION 2 Solo is equipped with two sequencing ports where PromethION flow cells can be run simlutaneously. The current chemistry and software configuration enables generation of 100 to 200 Gbases of data from each PromethION flow cell. Run times are 72 hours.
